I have seen different protocols abt leaf infection to deal with pathogenicity and control of plant pathogens especially when work focus on studying metabolomic and/ phenomics
Yet what are the performing critirea using only a leaf in the in vivo study especially when we know that there is more than the plant component that play the role in triggering pathogenicity and concluding/making decision in disease management strategies ?

Leaf assay for virulence /pathogenicity is a common method to test exactly virulence/pathogenicity. Look at LOPAT test for Pseudomonas differentiation....... You must use only susceptible variety or indicator plant with universal HR reaction (for bacteria or viruses). Inoculated plant must be grown at optimal conditions to prevent induced resistance.

I believe that leaf assay has a very important function, expecially when you are researching HR or intermediate resistance. I also think that leaf assay can quantify the virulence (ex. # spores x leaf area...) of a certain pathogen and isolate the capability of a certain plant to sustain the infection, excluding environmental conditions. Such studies are useful to detect new hybrids or prebreeding material and select then for field trials.

I believe that testing pathogenicity using leaf assay is very common and reliable methods especially if you are working with sensitive protocols that includes metabolic measurements to reduce the noise comming from different environmental factors.

It is important to determine the pathogenicity for plant pathogen causing leaf diseases, you can do it by two methods:
1. Injection the bacterial cells or fungal spores
2. Direct on the surface of a leaf by putting a part of media by cork porer

The assay you use depends on the question you are asking. For fungal pathogens, I don't like using detached leaf assays because the leaf is under stress and may not behave the way it would when attached to a plant, and if tissue age is an important factor, you may be adding uncertainty to the assay unless you use leaves that are exactly the same age. On the other hand, with detached leaves you can test many accessions simultaneously, quickly and inexpensively. I don't like wounding leaves because it makes it difficult to determine if the pathogen is a primary invader or a secondary invader. I don't like using agar plugs because some pathogens are good at infecting if they have a food base but poor at infecting when they don't. I like whole plant assays under controlled conditions, but I know that this also has its artifacts: If I infect a densely branched cultivar and a sparsely branched cultivar in a mist tent, they may show equal disease, but if I inoculated them in the field, the sparsely branched cultivar might show less disease because the dew dried more quickly. There is no bad assay, but for each assay you need to understand the assumptions you are making.

Obviously Detached leaves are interesting regarding highthrough-put analysis yet different parameters must be "controlled" at least at an extent
Another thing, we can orient our study using leaf test for pathogenicity and that depend on what profiles we need to observe
on the other hand the test is limited to "lab assay" therefore any bottelnecks must be considered at that point should be revealed and anayzed and that for sure will lead to more good answers about the limitation and advantages of this test
