I want to know that, what could be the best way to check (developmental progress) the mitosis and meiosis during spermatogenesis (in-vitro), Gene expression, immunohistochemistry or by some other method?

I did my PhD in this area (reproduction, Sox9, Sertoli cells).
Spermatocyts need a special environment for maturing and cells providing it and nurturing them. These 'nurse cells' are Sertoli cells which are known for the tightest tight-junctions they form to ensure the enclosed special environment. Blood-testis barrier (comparable with blood-brain barrier).
As far as I know, the only possibilities would be to take slices of testis or whole gonads in culture for several days, to do such thing in vitro. But then you need anyway the organism as donor for the gonads - so why not then let it happen in vivo?
To check for markers, take any paper investigating spermatocytes in different stages using immuno. Because that is what we did, too. e.g. in http://www.ijdb.ehu.es/web/paper/092874gk/dicer-is-required-for-sertoli-cell-function-and-survival-
