I would like to evaluate the in vitro effect of some new synthesized compounds with pharmacological potential on human cell lines. I would like to assess the anti-proliferative or anti-inflammatory effect of these compounds. Can anyone advise me regarding how to design this experiment? How can I determine the appropriate working concentrations? I would appreciate any advice/opinion.
I assume that you have at least an idea about the target concentration at which you would like to see a pharmacological effect, otherwise just as a golden rule, it would be difficult to develop a drug that has an IC50 or EC50 greater than 20micromolar (for a molecule of MW
First you should determine the effective concentrations for your drugs on cell lines.
Prepare working solutions of the drugs with concentrations of 100 nM, 1 uM, 10 uM and 100 uM. Then perform anti-inflammatory or anti-proliferative assay and eventually calculate IC50 values using linear regression model (use logarithm of the concentrations).
I agree with answers of Mr bondi and Miadvski. t is a matter of trial and errors. Another method you can start with. low drug concentration and at the same time try try another set with 10 times higher concentrations. Then go up or down till you hit your target. Good luck
