As mentioned by you, every Mass spec technology that is currently available in the market has certain limitations.Recent Orbitrap technologies such as Fusion and Fusion Lumos with multiple fragmentation features (CID, HCD, ETD, EThCD, ISD) seem to be much more competitive and near complete in terms of Protein/Proteomic related applications (Bottom Up, Top-Down, Middle Down). Recent publications suggest that Orbitrap can be used for targeted quant (PRM) analogous to TQ applications (SRMs, MRMs)

As pointed out by Hayakawa, Mass spec cannot give absolute quantification, nevertheless it can be confidently used for relative quantification approaches.

Triple Quad (TQ) can be used for targeted applications but they aren't fit for discovery or global Proteomic approaches.

Q-TOF technology is good but it lacks multiple fragmentation options which is very essential for in-depth characterization of proteins or large bio-molecules. Also, the proteome coverage obtained using Q-TOF is most times not comparable to that of Orbitrap' output...

MALDI deserves a place as it complements ESI based MS instruments and are much more robust but less sensitive as compared to their ESI counterparts.

Therefore, I would say Orbitrap would satisfy most of your needs related to Proteomic applications..

Dear Kou Hayakawa thanks for your reply but here complete  Protein Characterization is my concern.

Sravanan maximum i am getting positive feed back for Orbitrap technology which can make maximum characterization of all peptides with high resolution so in recent Bio-Pharma markets which are working on Monoclonal antibodies preferring Q-TOF for Intact mass analysis because of TOF technology you can go even Megadalton proteins,ADC and orbitrap technology for new protein Bottom-up sequence coverage where suits for Protein or Proteome studies because of its high resolution in 1HZ-5HZ.I am going to ask directly Bruker,Thermo and Waters application specialists for further discussion about there instruments. i will update soon.
