I am trying to pack 1 customised hydrogel bead in 1 drop using microfluid, yet the bead ends up outside the drop.
The customised bead (diameter 32um) also made from microfluid. The packing mf chip is with 80 um diameter. the flow rate is 15ul/min for bead and 23.7ul/min for 1.5% 008-fluorosurfactant, giving out drops with diameter ~110um. The concentration of bead is 10000/ml. Considering ideally it is 15ul/(4/3pi*55um^3)=21253 drop/min, I thought ideally I can get 50% drop successfully pack with bead. What could be the problems?

One reason may be the bead is quite big in relative to the drop and increase the interfacial tension. Will adding surfactant like IGEPAL CA 630 together in the bead mix reduce the interfacial tension and help with this problem?
