There is a difference between extracting polyphenols only for in vitro tests and in vivo or animal ones. For extracting and evaluation of the content only, we use generally methanol or ethanol at 80%, 70% and 50% with water (the three most polar solvent). For animal tests, it's better to use only water maceration by heating as decoction or infusion. The evaporation of solvent for in vivo tests is obligatory, otherwise the activity could be due to solvent, not to your plant.
If you can use the microwave, it's the best one (house microwave) and you can use a water as solvent in order to testing the extract without any effect on your animal model.
Hello, from my experience, the methanol (80%) is the solvent you can use. Since you work on animals, you can use water ou ethanol which are without effect (toxicity) on your animals.
If you want to get good extraction yield you should choose the solvent based on the polarity of target compound(s) that you are going to extract. You can get different degree of polarity by mixing different solvents such as water and ethanol at different mixing ratio.
