It depends a lot on the antibody, TBS (0.1M TRIS, 0.15M NaCL pH 7.5) is supposed to be a better buffer and is easier to prepare. PBS is based on NaPO4. There are several receipes for both.

As many scientists as many opinions... Means the westen blot should be set-up for every problematic protein/antibody from the beginning....
