Immortalized cells stop growing after incorporating a large plasmid?

I have been working on generating a stable CRISPRi cell line in Fibroblast and Smooth muscle cells. The problem is the cells stop proliferating after several rounds of proliferation and loose expression of Cas9 (or lose resistance to selection marker). Anyone with suggestions to navigate around this?
