I'm using a shoot apex from 4 day old germinated rice seedlings as my primary explants. I immersed my wounded explants into bacterial suspension culture for 1/2 hour at OD=1.0. I observed that there are heavy bacterial overgrowth after the explants were co-cultivated for only 3 days. Does that mean I should reduce my immersion time or bacterial concentration? I believe that if the suspension culture is too concentrated, chances of bacterial overgrowth will be higher.
