I am practicing microinjection now using phenol red and trying to measure the bead size on the micrometer. Everytime I inject the phenol red into the minneral oil, the beads immedietly burst and I cannot measure their sizes. My colleagues at the lab have no clues on why this is happening. I tried to optimize the balance, injection pressure, on-timer, and changed my thin-walled 5 times, and still doing the same thing. I need to measure the size of the bead in order to compute the MO volume. My desired morpholino bead volume is 1nL, and I am aiming for a diameter of 0.12 millimeter per bead. At this point, I really dont know what is causing this issue, and I am scared to use my Morpholino fearing of this issue might occue over and loosing my sample and rendering my subjects dead.

Can you please, kindly, and appreciatively help me or direct me on what to do or how to tacle this issue, or advise me if I am doing something wrong. Again, thank you very much for you help, I truly and sincerely appreciate it.

Sincerely,

Omar Issa,
