IHC Andy Algorithm, how to use it?

Hello everyone!

Probably, this issue was already raised, but I would really appreciate online help from someone who didi it. So - could anyone help me with fiji, Andy's algorithm? I try to run it (the tutorial now but I don't know about the calibration, don't know which treshold is best to use... problems, problems...:( Would be very thankful!
