I stimulate the human PBMC with CpG for 6 hours and use Brefeldin A for inhibiting the IFN-alpha secretion. Then I just use the regular intracellular staining. I found that the Brefeldin A is better than Monensin at this case.
I've successfully stained for IFN-gamma in CD8+ T cells. Not sure if that would translate, but it may be worth a try.
I used the Foxp3 / Transcription Factor Staining Buffer Set from eBioscience to Fix/Perm activated splenocytes. Let me know if this is of interest to you.
