Thank Dr Mostafa Haghpanah for your answer. I am afraid that adding EMS using filter at laminar hood will do harm to health if the volatile EMS out ot hood.
You can do the filtration in the fume hood. You can to avoid contamination if you use a sterile container on which the filter can fully fit. For the fear of expose, you have to work fast but very carefully, after all you will still work in the hood when using the chemical on your cultures.
Thank you, David. The explants I will use is from straberry growing in the field. EMS treat will be followed by surface sterilization with EtOH and HgCl2. Therefore, EMS need not be filtered. Is IT ok? Besides, hume hood is not sterile enviroment different laminar flow cabnet.
EMS along with 1% (v/v) DMSO as a carrier agent was prepared in deionized water and diluted with 0.1 M phosphate buffer (pH=7) using filter sterilization under aseptic condition.
