I'd prefer not buying any separate commercial media because of the cost. Please suggest what growth factors or vitamins can I add to DMEM KO other than glutamax, penstrep and bFGF for the MSCs to grow normally.

you can ev-deplete your FBS with ultracentrifugation - there are protocols. or you could use BSA to supplement. or you could see if the cells will tolerate being serum-free if it is for a short time.
Thank you! However, I don't have a facility to ultracentrifuge FBS in a sterile condition since I need EVs for functional assays. About BSA, I found several issues like lack of growth factors, lack of solid evidence to use it, plus I'm a little short of time to standardize it. Are there any other alternatives?
invitrogen does sell exosome depleted media, it's expensive though. i would recuomment just putting it on for exosome collect. washing cells with DMEM 3-4 times before adding it.
