Hi,

So am challenged with the parasite rescue transformation assay of Leishmania. I lyse the infected macrophages for 30 secs with 0.05% SDS but the parasites are not rescued. I was able to demonstrate parasitemia by microscopy, but after 48h of incubation following lysis, I do not obtain amastigotes in culture. Could someone please suggest a better alternative or add-on to assist me?
