Do you want to germinate and grow in the dark, or place light grown seedlings in the dark for induction? What is the aim?

In fact, I want to do the dark treatments of seedlings on MS plates with sugar and dark treatments of seedlings Also on MS plates with ABA. that's to study the phenotype by either sugars or ABA. Also, I want to measure Fv/Fm primarily. How can I do that please? I want to apply the dark on the seedling which germinates on MS media

On plates it will be easy. You can germinate in the light and after set times wrap in alufoil and place these wrapped plates in a light tight box (better to be safe). This also enable you to remove the plates on set times so you can perform different duration of the dark treatment. For dark germination you directly wrap the plates in alufoil and place in a box.

 For dark treatment usually, we wrap the plates with aluminum foil for the required days.

I would like to add a general point to the above mentioned protocols.

For germination and subsequent growth experiments it is essential that the developmental programm of the seeds is insync. To acchieve this, I usually put the prepared plates (already wrapped in aluminium foil) for at least 48h to 4°C. Then, at the start of the experiment, I always put the plates for 4 hours to light (best in a light chamber for even light quality), wrap them again afterwards and put them to the growth chamber. The light pulse of 4 hours starts the germination program in all vernalized seeds at the same time. With this you minimize biological variation what results in smaller error bars.
