I'm trying to do karyotyptic at different passages of the MSCs to evaluate the genomic stability during the culturing period as well as during differentiation pathway. I followed the protocol published for mononuclear cells with PHA, colchine and fixing and staining. though i could see swollen cells under microscope, I couldn't find a single bursted cells with chromosomes. Can anyone suggest me an appropriate protocol for stem cells to do the same.
