I want to knock down few genes in barley, I have designed gRNAs for them. Now there is the issue of plasmid selection that could be suitable for Agrobacterium mediated gene transfer.
From Addgene few plasmids are available that work for plants. Plasmids having binary function i.e they express both gRNA and Cas 9 simultaneously. I am not sure will they be working for barley or not.

There is a published version of the CRISPR-Cas9 that has been optimized for rice (sorry, I don't recall the reference, but you should be able to find it by searching). I have learned the hard way that constructs used in dicots often exhibit less than optimal (and sometimes zero) expression/function when used in monocots, and vice versa. I would make the effort to find the rice version before investing time, effort, and resources in a dicot version.
