can i put seed directly in bacteria which is grown in LB media, or maybe I have to make another suspension for seed treatment?

take 1 ml of fresh bacterial culture into a 500mL flask having 250mL LB media and shake in a rotatory shaker at 170 rpm at 28-degree centigrade for 48 hours. Now take 50 ml tubes and pour 48 ml from the culture that you just got from the rotary shaker. Centrifuge at 6000rpm for 15 minutes, you will get the pellet while discard supernatant. Then this pellet suspends in 100 ml sterilized distilled water. You can adjust the concentration of cells in suspension according to your need. Now take seeds, initially surface sterilized with 1 % NaOCL for 2 to 3 minutes and washed several time with distilled water. dry the seeds by placing them in sterile filter papers. Afterward soaks in the seeds in the suspension. again shake at 150 rpm at room temperature for several hours. The treated seeds air dry under LFU overnight. Now the seeds are ready for pot experiments.
