I've isolate secondary metabolites from marine fungus by extracting and fractioning the culture media of the fungus. But when i tried to subfractioning my methanol fraction, the fraction wouldn't separate. I've tried with UV-hplc reverse phase and water-methanol solvents. Any thoughts why my fraction wouldn't separate?
This is a very open ended question... with very little details
* What wavelength did you use to detect separation? What ever one you will use you will miss what ever does not absorb at this wavelength.
* What ration of MeOH to water did you use in the HPLC?
etc. etc...
Try to separate using MPLC ; Lsolera ONE;( Biotage) ( flash chrom.) on a reversed phase column using water and methanol.
flow rate of 20ml/min for the column and the detectin wavelength is at 254-366nm
