I use Fugene-3ul/well, pclEco- 0.8ng and polybrene 2ul/ml from 5mg/ml stock. I usually spinfect T cells that I have activated a day before with anti-CD28 and anti-CD3. I use MigR2 plasmid with my GOI in it.
To improve the efficiency of transduction, whenever you use the Polybrene agent as described, the most important thing is that the cells are over 40-50% confluence.

I suggest you first verify the virus titer before you transduction primary T cells. If the virus titer is very low, you need to confirm whether your Plat-E cell expresses gag-pol and env. In addition, polybrene agent is not essential for viral production, you can add polybrene agent in the transduction of T cells.
