I am looking for PCR primers that will amplify in the presence of any avian blood parasites infection which will indicate presence/absence of the infection in village chickens sampled in Nigeria. The primers should be able to amplify in the presence of Plasmodium, Haemoproteus, and/or Leucocytozoon. I wish to use PCR to identify all the infected samples, even at a low intensity infections that I might have missed while microscopically viewing blood smears. I would appreciate suggestion on any available published protocol that will serve as guide to my lab work as well as recommendations on which primers design will bring success to my work. The best way to go about it is to carry out a multiplex pcr where you can detect the for all the targeted parasite in a single pcr reaction. You can do some litreture search to find out whether someone has developed and optimized a multiplex pcr for poultry haemoprotozoans.
you should design the primers yourself. Find the identificatory sequence for those parasite species, the do an alignment and search for consensus sequence, then design the primer using that consensus as template. CLC genomic workbench is a tool that allows you to do this.
First of all you search the identificatory sequence for above said parasite species and then do the sequence alignment and search for consensus sequence, from that consensus sequence you design the primer by using available primer designig tools.
