Hi everyone, I am currently planning on starting an experiment with secretion samples which has a significant amount of endotoxin. However, I am planning on using these high endotoxin samples to treat human immune cell line. Does anyone know the extent to which endotoxin impact in THP-1 induced macrophage, raw 264 cell or other cell lines that are save from effect of LPS. Thanks
CD14 is the cell surface receptor for LPS. I have worked with THP-1 cells but have never looked at CD14 expression. I suspect THP-1 would be positive, since it is a monocytoid cell line. I do not know if anti-CD14 antibodies block LPS binding. Certain mouse strains - C3H/HeJ and C57Bl10 - are CD14 deficient and therefore LPS non-responsive [although low-responsive would be more accurate] but I am unaware of any human trait of LPS resistance/CD14 negativity.
