I have just started working on glucose dehydration to produce hydroxymethylfurfural (HMF) in only water. I am trying to quantify the amount of HMF produced using HPLC equipment (Elite LaChrom HITACHI ) with an ultraviolet detector and Alltech OA 1000 Organic acids column, using 5mM H2SO4 as a mobile phase under isocratic elution with a flow rate of 0.5 mL /min at ambient temperature. The maximum expected concentration from my reaction is 0.001M. However, after running the calibration curve under these conditions, the column got stuck and we have noticed an orange gluee substance coming out of the connections. Could it be the polimerized HMF?
My question is how to analyse the HMF in water using HPLC, what concentrations should be used and what is the sample pre-treatment that should be applied. I will be very grateful for any tips.
