Can someone please suggest how I induce diabetes in cell line specifically in keratinocytes (HaCaT cells) and appropriate bio-chemical test for detection?
Hello Amrita, I do not have experience with HaCaT cells, but nevertheless can immediately tell you that there is no way to make these cells to get diabetes. To get better advice from the ResearchGate community I would recommend you to refine your question (e.g., maybe you are interested to make the cells insulin resistant…?). Complex living organisms (e.g., human) get diabetes, not cells ;-)
Sir thanks for your quick response, I want to see the difference between non diabetic keratinocytes (HaCaT) and diabetic keratinocyte. Excluding the primary cell culture, is this possible?
You could try inducing insulin resistance through treating the cells with high levels of insulin for a period of time to downregulate insulin receptor, or through using something like IL1b, siRNAs for both the A and B insulin receptor subunits, or interfering in some way with the signalling pathway (i.e. MAPK, pten). You would have to test to confirm insulin resistance and these treatments will undoubtedly change the cells in other ways.
You have said you want to exclude primary cell culture but taking cells from diabetic/non-diabetic subjects (animal or human) might be easier and more relevant.
