The simplest approach is to create a knock "in" mutation for the gene and see it's effect on malignancy.

Also see, if you upregulate that perticular gene than what is the effect on tumor progression and malignancy...

Hi I agree with Ajay suggestion, that is the basic approach adapted. I would also recommend you to design a cell line in which you can conditionally over express the desired gene in response to any inducer (similar to tet off and tet on condition) This will almost mimic your in vivo situation.
