I have identified CD41+ CD42+  CD45+ human cells in culture, but would like to confirm these are truly megakaryocytes. I was hoping that I could fix the cells and use 7AAD as as a dye to show endoreduplication. I imagine that cells which are 4N or more would incorporate more 7ADD than 2N cells. Does anyone have experience with such a protocol?  Alternatively, what is the best method to show that we have platelets in culture?
