Hi,

I'm experimenting to check the effect of treating polyIC (poly(I:C), polyIC, poly-IC, polyI:C) to NS-SV-AC cell lines. But I haven't been witnessing a significant cell death after treating polyIC, unlike many references available online. Hence, I began to wonder if this is due to my lack of transfection skills, or if it's because I'm using the wrong dose, or if it's because I'm not keeping the incubation time right. It could also be that polyIC is not being transfected at all (I'm trying to "transfect" using both lipofectamine and P3000, along with opti-MEM, not "treat" polyIC.

If anyone's familiar with transfecting polyIC to cell lines, could you please share with me the know-how? About the ratio of mixes I have to prepare using lipofectamine and P3000, as well as the ideal incubation time?

I'm currently seeding 1x10^5 cells to 35mm dish, with 1 ml of media with 10% FGR and 1% AA.
