That really depends on the amount of total protein you load on your gel and the specificity of your primary antibody.

Let me first ask you the most fundamental question here.

Do you have a verified primary antibody against your protein of your interest?

I guess the issue is with primary antibody. It has been often seen that one antibody which is working for one cell line may not work for others like cancer cell lines. Better to try another antibody.
