I want to tag antibody with hydrazine to do bioorthogonal experiments, and followed by reacting it with aldehyde.
1. Are there any protocols for such reaction?
I found general bioorthogonal reactions involving aldehyde-tagged proteins with hydrazide targets. Suppose if I want to do it inversely, is it reasonable to use EDC/NHS to activate antibodies carboxylic groups in the presence of N2H4 for coupling?
2. Is there any problems with hydrazide-tagged antibodies and aldehyde based targets?
In theory, the terminal carboxilic groups in your antibody will form hydrazides, that will be able to react with aldehydes to form hydrazones.
Hydrazine could react with any carbonyl group (ketone and aldehyde), if your protein can have some of those groups (glycosides, etc) could interfere with your tagging.
