I am carrying out an experiment that nanoparticle synthesis by desolvation method. For the bonding i target, firstly i need to keep pH of the reaction at 8-10. I use bovine serum albumin. In literature, they add NaOH on BSA before desolvation. However, if i add NaOH before desolvation, the pH return to the neutral condition after addition of the solvent as expected so, i have to add NaOH after adding desolvating agent. If experiment is conducted in pure water (i mean if i dissolve BSA in pure water and i add desolvating agent on it) turbidity is completely loss after i add NaOH. In PBS, turbidity is not completely lost, but a small loss of turbidity is still observed and some of proteins aggregate.
Why this situations are observed? I couldn’t understand the effect of NaOH on protein and is there any advice to keep nanoparticle synthesis at basic conditions?
