Hello everyone;

I am trying to develop HEK293F suspension cells for recombinant protein production. I use the Expi293 Expression System kit for this. HEK293F cells were cultured in Expifectamine medium at 37 °C under 8% CO2 with agitation at 120 rpm (as 30 mL culture in 125 mL flask). Cells need to be 3–5 × 106 viable cells/mL to be subcultured. However, the cells grow until they reach a maximum of 1.6 x 106 viable cells/mL, then their viability decreases and they begin to die. I can't grow the cells to the number requested in the protocol so I can't subculture. Additionally, the live cells decreased in size over time and don't appear to be the size of normal HEK293F cells. What can I do?
