I need to store blood samples for some time (a couple of hours) to then perform Hoechst dye staining for flow cytometry analysis without having substantial loss of cells. Any suggestions or protocols that would allow me to do this?
Dear Gomez, Blood sample can be kept at 37oC for couple of hours as it is.
First question is, you are doing Hoechst staining in whole blood? (then you can keep it as it is for couple of hours)
then what are the cells of your interest? (if PBMC or any particular WBC, then do it immidiately after isolations of cells from blood, or keep in cell culture medium e.g. RPMI for couple of hrs).
