Hi, here my problem :

I want to see and measure the potential membrane of mitochondria in chondrocytes cells using the dye TMRM. In culture cell it's very easy to do and I don't have difficulties but I want to use it in situ. The sample must be cut in very fine ends to allow to see correctly the mitochondrias into the cells, so far I've seen in various protocoles the use of a microtome to do the cut but the sample must be fixed. I'd like to know if someone know a technics to process a sample and to obtain very fines end without the necessity of fixing the cells during the process which could allow me to use the TMRM dye.
