How to screen biocontrol agents(Mostly fungal) from the soil? I have to use these biocontrol agents against Rhizoctonia solani. If you have done any research on the screening of biocontrol agents, do share your papers or protocols with me.
RS mostly form the sclerotia and you can perform primary screening by direct confrontation assay. 2nd you need to use slcerotia mortality test by using different spore suspension of your potent fungal bio agents. Generally Chitinase producing fungal bio-agents are potential against RS.
We find that Trichoderma harzianum is a good bio control agent against Rhizoctonia solani under arid soils of India. However it has been reported that Greek indigenous Streptomycetes is a good bio control agent against RS. Please consult:
For specific isolation I would suggest baiting bag technique (using R. solani) where bait (sclerotia rapped in muslin cloth pouch) to be buried in the rhizosphere (desired sampling site), and it will attract the potential biocontrol organisms towards it and after few days you can collect pouch along with surrounding soil and go for isolation.
