How to screen a large collection of different bacterial isolates with lowest contamination risk (liquid LB medium)? and, Do you have any suggested how to keep them in -80 with limited space?
We will have a large collection of different bacteria together which we need to screen them. Although, I know that one of the possibility is keeping them in 96 well plates to not get too much space but anyone has any other suggestion? Even if we sealed them by stickers, we will have the risk of contamination. Do you have any other suggestion in this regards?

I suggest storing bacterial strains in the liquid media (depending on the taxonomic position) plus 15% glycerol, at - 80 C.
I think you may use 96-well micro-plates too. But, be careful to avoid cross contamination when you inoculate the wells.
It depends on how you screen the bacterial library. generally, you can obtain individual clones by plating the library on non-selective medium plates such as LB. Or sometimes you can screen by desired phenotypes such as products, resistance or pigments. If you want to store individual clones, you can store them on 96-well plates with 20% glycerol. If you store the mixed library, you can simply store them in 2-mL tubes with 20% glycerol.
the best is to use small glass capillaries as container for frozen isolates (20% glycerol or DMSO).
