I am going to purify an enzyme from serum by precipitation with ethanol but the problem is the lipids make the resulting precipitate insoluble in the buffer so how can I remove this lipids?
The method developed by Cham and Knowles (1976) J. Lipid Res 17, 176-181 works very well to remove the majority of serum or plasma lipids. We adapted this method for a microscale purification of gangliosides: Ladisch and Gillard (1985) Anal. Biochem. 146, 220-231. Of note, our partition method leaves about half of the GM3 ganglioside in the upper organic phase.
