I am trying to quantify the number of bacterial cells (e.g lactobacilli) in cow milk. I have extracted DNA from my standard (i.e lactobacilli). I have done a 1:10 dilution and i have done a standard curve from series of these dilution. I have also extracted DNA from my cow milk samples and i have ran this together with my standard on qPCR. Now i want to know how to calculate the concentration of bacteria in my sample and how to relate this back to the number of bacterial cells considering that the qPCR is only giving me the concentration of DNA in my sample. How do i know the actual bacterial number or cells in my milk sample. Thanks
