I am trying to purify a protein of interest from E coli periplasm. The protein is tagged with pelB signal peptide at N term and expressed very well. I tried to purify it using osmotic shock (Sucrose and MgCl2) but the protein remained in cells only. It did not come in solution. It seems to me that protein is soluble but osmotic shock did not work well. I would appreciate if anyone can suggest the consequences of adding Lysozyme and sonication to get the protein as soluble fractions.
