Dear friends i have only H&E stained slide and i have to do other special staining from this slide only. kindly suggest me the procedure for complete destaining of this slide.
I have never used a Tris buffer to remove hematoxylin, but there is no reason that it shouldn't. The protocol listed by Alexandr Chernov will absolutely work, but it is longer and more intricate that it needs to be.
Clinically, we de-stain H&E slides as a regular protocol using 1:1 Xylene/Acetone solution to remove the coverslip and 1% Acid (HCl) Alcohol to remove the stains.
Acid alcohol is used in regressive hematoxylin staining regularly and eosin is quickly removed by both acetone and alcohol. By far the longest step is waiting for the coverslip to come off which can take up to an hour if the media has fully cured.
