We would put agar plugs in 25% glycerol and stick them in the minus 80. I will say that 25% is not appropriate for all species. I would test a range between 10% and 25% to see what gives the best recovery rates.
Also can put agar plugs in sterile mineral oil. Store at room temperature. It is useful for many species and fungi remain well preserved for couple of years.
It depends a lot on the fungi you want to preserve, not all of them can withstand cryopreservation. In our laboratory we have used Fusarium cryopreservation in 20% glycerol and the sterile filter paper method for fungi such as Exserohilum and Bipolaris.
I would recommend using more than one preservation method and evaluate viability after a few days or weeks.
