1/ Solution preparation: How to prepare these solutions below

Your stocks are: 1M tris pH 8; 3M NaCl; 3M imidazole; 50% glycerol; 10X PBS

Prepare these solutions (Final concentrations are in parenthesis):

• 1X PBS,
• 10 ml of EQ: Equilibration buffer 0 (50mM Tris pH8, 150 mM NaCl, 5% glycerol,1X PBS)
• 10 ml of EQ5: Equilibration buffer 5 (50mM Tris pH8, 150 mM NaCl, 5% glycerol, 5 mM imidazole, 1X PBS),
• 5 ml of EQ15: Equilibration buffer 15 (50mM Tris pH8, 150 mM NaCl, 5% glycerol, 15 mM imidazole, 1X PBS),
• 2.5 ml of EQ250: Elution buffer (50mM Tris pH8, 150 mM NaCl, 10% glycerol, 250 mM imidazole, 1X PBS).
