Dear all, replicating a protocol from a paper I have to prepare a borate buffer to dilute poly-D-lysine for a coating. The paper shows this recipe:

Boric acid 100 mM

Sodium tetraborate 25 mM

Sodium chloride 75 mM

but I can't dissolve the powders properly (some granular remains persist).

Collaborators lent us their borate buffer which is concentrate to a half without NaCl, basically:

Boric acid 50 mM

Sodium tetraborate 12,5 mM

Since during washes for fixation I have issues with cells detaching I am trying to find a way to stay more strictly in line with what the paper says.

Is there anyone with advices on why I cannot dissolve the powders at the higher concentration? Anyone with experience with this buffer? Is there any trick like for the dissolution of EDTA where it is not soluble until you increase the pH with NaOH?
