I have isolated about 20 different kinds of Fungals from the fruit peel and now I would like to measure the spore concentration,
1- How can prepare the spore suspension solution for spore counting, as not sure which kind of fungi?

Harvesting of spores was carried out from the surface of solid media by pouring 5 ml
of sterile distilled water containing 0.05% Tween-80 into the fungal plate. It was then
scrapped by using scalpel for about 2 min. It was then filtered using muslin cloth and filtrate
was used as stock for spore solution.

First, prepare your fungal inoculum in solid agar medium or liquid medium.
and then harvest your spore by filtering (to sperate the spores from hyphal freagments) the culture liquid medium or scrap the fungal surface colony with the spreader (from solid agar medium). your spore suspension then ready to calculate your sporea using haemacytometer
