For this, should the culture broth is to be separated from the cells, then the media should given directly with the addition of HNO3 or should broth has to be autoclave and then given for analysis.
Depends on what you want the answer for: if its only the concentration in the broth, then you remove the cells first, otherwise you dont get a differenciation between whats inside the cells and whats still remaining in the broth. In any case you should subject the liquid sample to a HNO3/microvave digestion before AAS.

Many different kinds of samples from diverse industries can be analyzed by using either FAAS and GFAAS following simple preparation procedures. A typical sample preparation procedure for solid and viscous liquid samples involves digestion with a concentrated acid; for example, HNO3, HCl, or H2SO4. After dilution of the digested solutions, samples can be directly injected into flame AAS as well as graphite furnace AAS. Exactly 3g of sample was weighed into a dry crucible and ashed at 500oC for 30 minutes in a muffle furnace. The ash obtained was dissolved in 100 mL aqua-regia and the content heated to dryness without baking. The digest was dissolved with glass distilled water and the volume made up to 50mL with glass distilled water and stored in a clean plastic bottle for analysis.
