I am preparing Rifampicin (injectable) with a final required concentration of 5μg/5μL by dissolving a 450mg tablet in 100% DMSO. I want to inject 5μL of the drug solution into my animal model. The solubility of Rifampicin in DMSO has been provided as 100mg/mL (Rifampicin(sigmaaldrich.com)).
We dissolved the tablet in 4.5 mL of 100% DMSO, and a volume of 10μL was diluted to 10mL (so that the DMSO gets diluted to 0.1%). This makes my final solution have a concentration of 0.1mg/mL. This final concentration does not provide my final required concentration of 5μg/5μL. What can I do in this situation to achieve my final, 5μg/5μL, concentration? Can someone provide a valuable suggestion?

Rifampicin is too hydrophobic to reach 1 mg/mL in an aqueous buffer even using DMSO 0.1%. We developed a rifampicin formulation by encapsulation in PLA nanoparticles, in an aqueous buffer, and we can reach 4 mg/mL. If interested, have a look on www.adjuvatis.com and contact me for more information.
