I am new PhD student and I have plan to start an experiment which is requires to do different techniques. Therefore, I would like any researcher find my post to answer my question or give me any suggestions:
1-Can anyone help me to get a proper protocol for preparing conditioned media after transiently transfecting with specific plasmid, because I am planning to test the effect of other cells with this medium?
2- Is there any suggestion on how to test the the transfected gene on this condition media?

What type of cells are you planning to transfect? And what is the product that you expect from the transfection?

First thank you for your replay,
I want to transfect colorectal cancer cells (HCT116) that is low expressor with the gene of interest. Then I want to collect the conditioned media after transfection to test the effect of this media on another cells (primary cells) through performing different functional assays (migration , invasion, proliferation).
