For drug release study, you need to optimize the parameters such as temperature (usually 37C) and pH etc. Also its important to know whether the drug is hydrophilic or hydrophobic, since hydrophobic drug will not easily dissolve in water or pbs.

Then, you need to take readings at regular time intervals and add up the concentration to get cumulative drug release profile.

Try using magnetic stirring if the particles are being clumped and not releasing the drug.

For drug release study you have to make standard curve of pure drug by using UV-VIS spectrophotometry, from where you can compare the release profile of drug loaded nanoprticles at regular time interval.

from where drug concentration can be calculated by Y =mX +/- C and

Amount of drug = Concentration × Dissolution bath volume × dilution factor

Although UV vis kind of a standard technique used in lab-based drug release to be super precise you should opt for HPLC, that should be able to give you. It also depends when you are saying satisfying results. what is the definition of satifying results for you? The :) Release profile can be a burst or sustained release. how long did u kep the experiment running, You can alway use flow cytometry to get a real time release profile as well. Chitosan is polycationic in nature what is the charge on catrotenoid:: is it a covalent or ionic bond. A loading concentration and entrapment percentage in nanoparticles as well is critical. What concentration are you loading these nanoparticles at, that depends on the minimum inhibitory concentration of your targetted disease or problem? So a lot of factors play a critical role. The question needs to be more specific. Hope this helps.

Thanks a lot sir Saadi Bin Qasim for your suggestions. Sir i have performed dialysis process for 7 days and analysed the PBS by UV-spectrophotometric method. I am getting very low concentration of drug in the buffer and it is negligible if it is compared with the initial concentration of drug present in CNPs. That is why I am not satisfied with my result. This dialysis method is reported in most of the research papers. Specifically I want to know is this method is appropriate or not for my purpose?? Or i need to modify any step or follow any other method? Thank you.

What concentration are you loading the drug at. Whats the charge on drug. you think its binding with the chitosan nanoparticles, It might be a strong covalent interaction . People have also used a USP Paddle, setup for dissolution/drug release as well.

I think that you need to check the solubility of the carotenoid pigment. "Most carotenoids are insoluble in water and are soluble in organic solvents such as acetone, alcohol, THF, ethyl ether, chloroform and ethyl acetate" - Rodriguez-Amaya, D.B. A Guide to Carotenoid Analysis in Foods; International Life Sciences Inst: Washington, DC, USA, 1999. It is also important to determine the saturation concentration of carotenoid in the bulk medium - i.e the max that can dissolve in the specific volume you're using. Choosing an appropriate solvent is important, as well as an appropriate analytical technique for quantification of the amount of drug released. It is also important to consider the volume of bulk/receptor medium and the amount of sample (with a known carotenoid concentration) that you will use. The nanoparticles could be clumping together due to a poor aqueous solubility. You may need to use an appropriate co-solvent or a surfactant but this may influence: the pH of your bulk medium, the integrity of your nanoparticles and the release profile.
