Good day, I have to perform a viral load analysis for some samples for HIV-1. I know how to make a viral stock of HIV-1; however I have no experience in viral load analysis, which i hope i can find some insight. These samples will derive from a bank of virus that i will make. Afterwards, i have to infect my cells with a specific number of multiplicity of infection (MOI). However, to achieve this MOI, i have to perform a viral load analysis to know how much particle of virus i have per mL.
Does someone have experience in performing viral load analysis or know how to?
Is viral load analysis done by RT-PCR procedures?
Can someone explain to me multiplicity of infection and how this correlates with viral load?

For quantitative NASBA-based viral load assays
using homogeneous detection with molecular
beacons, such as the NucliSens EasyQ HIV-1 assay,
a quantitation algorithm is required. During the
amplification process there is a constant growth in
the concentration of amplicons to which the beacon
can bind while generating a fluorescence signal.
