Hello everyone, I need some help with the Nanodrop 2000c for dosing RNA concentration of my samples. It's the first experience for me with the nanodrop when it comes for RNA measurement, so I extract my viral RNA with QIAGEN QIamp Viral RNA mini Kit, and after the extraction, I get good concentration and good ratio on 260/280 but a very poor ratio on 260/230 even a negative ratio. So I don't know if this information is reliable or do I need to improve my extraction procedure ? and how ? and what if the measurement will be affected if the sample was in -20°C for a long time ?
Also, is it possible to measure my cDNA and do I need to select Single or Double Strand-DNA on the software ?
